NUCYNTA™ Launches in the United States: Milestone for Pain Franchise
We are proud to announce today's launch by PriCara® of NUCYNTA™ (tapentadol) CII immediate release tablets, the first new centrally acting analgesic in more than 25 years. Congratulations to the many people across Johnson & Johnson Pharmaceutical Research and Development, PriCara, and other Johnson & Johnson operating companies who have worked tirelessly toward this goal for many years. Your efforts will make a difference in the treatment of acute pain and in the success of our pain franchise, and you are to be commended.

NUCYNTA is indicated for the relief of moderate to severe acute pain in patients 18 years of age or older. It is classified as a schedule II controlled substance. This treatment represents a new option both for patients with moderate to severe acute pain, many of whom are dissatisfied with existing therapies, and for health care professionals who treat pain.

NUCYNTA has a different approach to pain relief. It is a single molecule with two mechanisms of action. It binds to mu-opioid receptors and inhibits norepinephrine re-uptake. Although the exact mechanism of action is not know, these two mechanisms, which affect established pain pathways, are thought to be responsible for pain relief with NUCYNTA.

In Phase 3 clinical studies, NUCYNTA demonstrated comparable efficacy to the current standard of care for acute pain. It was found to be safe and effective in multiple pain models, and it is well tolerated, particularly in regard to gastrointestinal side effects. Clinical trials showed NUCYNTA had relatively low rates of discontinuation due to adverse events, which is a challenge with other medicines in its class.

Pain is a serious medical condition that often goes undertreated. Acute pain, in particular, may cause considerable medical complications and impair patient recovery. Each year, millions of Americans experience acute pain, which is the most common reason people seek medical attention. NUCYNTA has the potential to improve treatment outcomes for millions of acute pain sufferers.

We are dedicated to marketing NUCYNTA in a responsible manner to our customers. We will uphold this commitment by continuing to sponsor and support educational programs about safe and responsible use of pain medicines and prevention of inappropriate use.

I encourage you to learn more about NUCYNTA by going to www.pricara.com or www.jnjprd.com to review the company press release and full prescribing information for NUCYNTA.

We trust that you will share in the excitement of the day and feel proud of this latest milestone in our pain franchise.
